Preparation of the Research Cell Bank
One vial of the frozen high ХХХХ expression transfected pool of modified CHO cells was used to prepare the research cell bank.
The contents of the vial were revived in the chemically defined suspension medium in T-175 Flask.
The T-175 flask was then incubated at 37°C and 5% CO2.
Once enough cell count was achieved the flask was expanded into a roller bottle in the same medium and kept in roll-in incubator at 37°C, 5% CO2 and 10 rpm.
The incubation was continued till the cells reached the log phase of growth.
At that point, the cell suspension was centrifuged and then re-suspended in the cryo-preservation medium.
The suspended cells were then aliquoted into cryo vials and then frozen at -180oC and stored at the same temperature.
