what do you want know or do?

Infact it depends on the context. It means what do you want to know more about this matter for someone wants to know more about something unknown to him.
Or as the other answers that what do you want to do in the future?

What's going on in your life lately?

You can answer anything you'd like, from what you are doing right then to what you've been "up to" professionally, or what has been going on in your life personally, or even what you're thinking about that moment. It's related to the Bugs Bunny question, "What's up, doc?"
