Source text - English Heat shock proteins (HSP) are molecular chaperones that assist in the structural formation and folding of a wide variety of client proteins such as HER2, ER, N-RAS, AKT,C-KIT, PDGFR, and B-RAF. Those proteins that are misfolded become subject to ubiquitination and proteasomal degradation. HSP90 is the most abundant molecular chaperone and is essential for cell survival.
AUY922 is an isoxazole that competitively inhibits the ATPase activity of HSP90. In doing so, the client proteins of HSP90 begin to undergo cellular degradation and the regulation of cell survival, proliferation and apoptosis are therefore affected. Generally, cells under stress produce and/or depend upon higher levels of HSP90 compared to non-stressed cells. HSP90 is an important target for cancer chemotherapeutics because tumor cells, especially those with mutations, exist in a stressful environment and depend
on HSP90 to grow and survive. For this reason, HSP90 inhibitors such as AUY922 are considered to be agents with significant therapeutic potential in a wide variety of tumor types.
Translation - Italian Le proteine da shock termico (HSP) sono chaperon molecolari che assistono la formazione molecolare e i processi di folding (ripiegamento) di una vasta gamma di proteine client quali HER2, ER, N-RAS, AKT,C-KIT, PDGFR, e B-RAF. Le proteine che subiscono un misfolding (errato ripiegamento) sono soggette a ubiquitinazione e degradazione del proteosoma. L’HSP90 è lo chaperon molecolare più importante ed è essenziale per la sopravvivenza della cellula.
L’AUY922 è un isossazolo che inibisce competitivamente l’attività ATPasica dell’HSP90. In tal modo le proteine client dell’HSP90 vanno incontro a degradazione cellulare, con conseguente compromissione dei processi di regolazione della sopravvivenza e della proliferazione cellulare e dell’apoptosi.
Generalmente le cellule stressate producono e/o dipendono da livelli maggiori di HSP90 rispetto alle cellule non stressate. L’HSP90 è un target importante per le terapie chemioterapiche antitumorali poiché le cellule tumorali soprattutto quelle che presentano mutazioni, sopravvivono in un ambiente stressato e necessitano dell’HSP90 per crescere e sopravvivere. É questa la ragione per cui gli inibitori dell’HSP90 quali l’AUY922 sono considerati agenti dotati di potenziale terapeutico importante per molti tipi di tumori.
