Source text - English Ozone (O3) is a naturally occurring
atmospheric gas composed of three oxygen
atoms; it is highly unstable (rapidly decomposing
to normal oxygen, O2) and is a
strong oxidant.
The therapeutic potential of O3 is
known since the end of the 18th century but
its medical application has been limited for
a long time due to the doubts about its possible
toxicity. However, in the last two
decades the medical use of O3 has been progressively
increasing all over the world as
an alternative/adjuvant treatment for several
diseases.1-3 Some mechanistic evidence has
recently been provided for the dose-dependent
effects of O3 treatment.4-6 In particular,
it has been demonstrated that low O3 concentrations,
by inducing the so-called
eustress,7 do activate cell anti-oxidative
response pathways8 which are likely
responsible for the therapeutic effects
observed in the clinical practice.
However, the potential of mild ozonisation
in tissue regeneration and differentiation
has been scarcely explored so far. In
this view, we investigated the effects of low
O3 concentrations on human adiposederived
adult stem (hADAS) cells i.e., the
mesenchymal stem cells which are present
in the stromal-vascular fraction of the fat
tissue. hADAS cells are able to differentiate
in vitro into meso-, ecto- and endodermal
cell lineages and can also be reprogrammed
as pluripotent stem cells more efficiently
than other cell types.9 hADAS cells are
therefore considered as a powerful tool in
regenerative medicine and tissue engineering.
9-13 In this study, we induced hADAS
cells to differentiate into the adipoblastic
lineage, and investigated the effect of mild
ozonisation on the adipogenic process; histochemical
and morphometric analyses at
light microscopy were combined with ultrastructural
morphology at transmission electron
microscopy (TEM).
Translation - Portuguese O ozônio (O3) é um gás atmosférico de ocorrência natural composto por três (3) átomos de oxigênio. É altamente instável (decompõe-se rapidamente a ‘oxigênio normal’ (O2)) e é um poderoso oxidante.
O potencial terapêutico do ozônio é conhecido desde o final do século XVIII, porém seu uso médico tem sido limitado por muito tempo devido a dúvidas sobre sua possível toxicidade. Entretanto, nas últimas duas décadas o uso médico do ozônio tem aumentado progressivamente em todo o mundo como um tratamento alternativo/coadjuvante para diversa doenças. 1-3 Algumas evidencias mecanicistas tem sido recentemente fornecidas para tratamentos com ozônio com efeito dose-dependente. 4-6Em particular, tem sido demonstrado que baixas concentrações de O3 ativam a resposta antioxidante das células por meio do conhecido euestresse 7. Tais respostas a baixas concentrações de O3 assumem a possível responsabilidade pelos efeitos terapêuticos observados na pratica clínica.
Entretanto, o potencial da ozonização moderada na regeneração e diferenciação tissular tem sido pouco explorada até agora. Levando- se em conta esta abordagem, investigamos os efeitos de baixas concentrações de ozônio nas células hADAS, ou seja, as células-tronco mesenquimatosas as quais estão presentes na fração estromal-vascular do tecido adiposo. As células hADAS podem diferenciar-se in vitro em linhagens de células mesodérmicas, ectodérmicas e endodérmicas e também podem ser reprogramadas como células- tronco pluripotentes de modo mais eficiente que outros tipos de células.9 Ditas células (hADAS), portanto, são consideradas como uma ferramenta poderosa na medicina regenerativa e na engenharia de tecidos. 9-13 Neste estudo, induzimos células hADAS a diferenciarem-se na linhagem adipoblástica e investigamos os efeitos de uma ozonização moderada no processo adipogênico. Análises histoquímica e morfométrica feitas com microscópio óptico foram combinadas com morfologia ultra estrutural feitas em um microscópio de transmissão de elétrons (TEM).
