Source text - English The ability of flurbiprofen to induce DNA repair (unscheduled DNA synthesis; UDS) as an
index of genotoxicity was assessed in hepatocytes isolated from male Fischer 344 rats, the
strain used in the second carcinogenicity study (TX 84036). Flurbiprofen had no effect on
UDS at concentrations of 0.1 to 100 μg/ml, which was considered to be an adequate dosing
range, as flurbiprofen was cytotoxic at the top dose of 100 μg/ml.
In the Ames assay, S. typhimurium strains TA 98, 100, 1535, 1537 and 1538, were exposed
to doses of flurbiprofen up to 2,000 mg/plate, with and without metabolic activation using the
9000 x g supernatant from liver of Arochlor 1254-treated male rats. Positive controls were
variously 2-acetylaminofluorene (for TA 98, 100 and 1538), cyclophosphamide (for TA 1535)
and 9-aminoacridine (for TA 1537). Flurbiprofen demonstrated no mutagenic effect at
concentrations up to 2000 mg/plate, with or without metabolic activation (PR88007).
Flurbiprofen’s potential genotoxicity was evaluated in an in vivo rat micronucleus assay. CD
Rats were administered gavage doses of 250, 500, and 1,000 mg/kg/day flurbiprofen at
0 hours and 24 hours. Bone marrow samples were collected 48 hours after the initial dose.
The results showed no significant increase in micronuclei in bone marrow cells from rats at
any dose level (30174).
In all four studies, flurbiprofen was negative for genotoxicity, and provides adequate
substantiation to indicate that this drug is unlikely to pose a risk of genotoxicity among
humans exposed to this drug. In addition, flurbiprofen is a phenylalkynoic acid, and this
chemical structure does not contain functional groups that might be “structural alerts” for
genotoxicity (Ashby and Tennant et al., 1989; Sawatari et al., 2001), nor are “profen”
NSAIDs considered to be mutagenic or genotoxic hazards (as evidenced by Table 1 of
Brambilla and Martelli, 2009).

Результаты не показали значительного увеличения микроядер в клетках костного мозга крыс при любой дозе (30 174).
Во всех четырех исследованиях флурбипрофен не оказался генотоксичным, что обоснованно указывает на маловероятный риск генотоксичности этого препарата для людей. Кроме того, флурбипрофен представляет собой производное фенилалкановой кислоты, и его химическая структура не содержит функциональных групп, которые могли бы быть «структурным признаком токсичности» в плане мутагенности и генотоксичности (Ashby and Tennant и др., 1989; Sawatari и др., 2001). Также, "профен" НПВП, абсолютно не представляет мутагенной или генотоксичной опасностей (доказано в таблице 1, Brambilla and Martelli, 2009).
