Senescence-associated B-Galactosidase Activity. A cellular senescence marker assay was performed as described by XXX. Briefly, cells were fixed with 2% formaldehyde/0.2% glutaraldehyde in PBS and then exposed to 1 mg/ml X-Gal in an iron buffer at pH 6. Dark blue precipitate forms in older cells proportionally to approaching senescence. Replicate wells were also stained at pH 7 as a negative control to show specificity of activity, and care was taken to avoid artifacts caused by overconfluent or quiescent cultures. Positively staining cells were expressed as percentage of total cells in independent wells.
