in the absence of further context it looks to me as if the "en" might mean "the 'copartes' introduced defenders (in)to their rights" i.e. familiarized the defenders with their rights so the defenders could spread this knowledge amongst the villagers (?)

Regarding the preposition it should be "promotores de..."

... depending on whether that is these people's function according to the rest of your context. "rights defenders" is found more frequently than "rights promoters" in a Yahoo search (787,000 hits compared with 1,590).

Here is a description of what these people do (please see link): "Se formó inicialmente con 142 promotoras capacitadas y trabajando por la promoción y defensa de los Derechos Humanos, con la misión de popularizar el conocimiento de los Derechos Humanos así como su defensa práctica."

neutral Marco Ramón : defenders, promoters, advocates. One NGO calls itself HRDefenders and Promoters. So there are all these degrees of "closeness " to the term.
