I did a quick Google and it appears it means "group" "combi" "ensemble" - that sort of connotation. A slightly more lofty way to say "group."

For instance, I could imagine that the Beatles "Fab Four" might use this word in a theoretical Japanese translation.

You should not treat the word 人衆 from the number (of people) that preceeds no matter what the number is .... i.e. 3 人衆, 4人衆, etc. In this case 5人衆 is one word, no more no less.
Also it is not just a group that happen to be there put together. The formation of the group is rather an act manifested out of desire or willingness to be so.
It is something like the old movie "The Magnificient Seven" (of course in this case # of people involved are five).
