At least five different amounts of test chemical are tested, with half-log intervals between plates. Substances are tested up to the limit of solubility or toxicity. Toxicity is evidenced by a reduction in the number of spontaneous revertants, a clearing of the background lawn, or by degree of survival of treated cultures. Non-toxic chemicals should be tested to 5 mg per plate before considering the test substance negative.

as for the previous question...

Bacteria are plated out with the test agent (perhaps in top agar as before, or perhaps spread over the surface of the nutrient plates). In the control plates (no agent) the bacteria will grow and will form a "lawn" - a homogeneous growth over the plate (this appears as a cloudy, sometimes "fluffy" surface. Depending on the test agent, growth will be eggected and so growth will be less (less cloudy lawn) or more (more cloudy lawn). At a certain concentration, all growth will be killed of and so the lawn will be cleared.

In other tests, plates with an already grown bacterial lawn have disks soaked in agents placed on them and their effect on growth will result in clear patches developing on the plate die to the bacteria being killed off (i.e. the lawn being cleared).
