First, structural detail that is not uniform throughout the thickness of the section will not be well resolved. This problem arises whenever the structures present are smaller than the thickness of the section, e.g. the nm-scale envelope surrounding cellulose microfibrils. This can be minimised by examining parts of sections perpendicular to the plane of the cell wall with the microfibrils viewed end-on.
